条件一致才能比较
Protein A 亲和层析是单克隆抗体捕获工艺的标准,但填料价格昂贵、寿命有限这一点一直被视为课题。[7][13]已公开的结合载量所用的抗体和保留时间各不相同,仅把数字并排放在一起无法构成比较。[1][5]
动态结合载量
动态结合载量(dynamic binding capacity,DBC)是在实际流动上样的条件下填料所捕获的抗体量。该值在很大程度上取决于保留时间(residence time),也因抗体种类和填料而异。[1][2]通常以上样至流出液 UV 信号达到最大值的 10% 的时间点(10% 穿透)为止的上样量来计算。[4][5]因此,应在保留时间和穿透标准相同、并且尽可能用本企业料液测得的数值之间进行比较。
基质与粒径
基质(base matrix)分为玻璃·硅胶类、琼脂糖类和合成聚合物类。[1]在一项比较 15 种市售填料的研究中,对保留时间的依赖性因基质而异,该研究以颗粒直径和传质速率解释了这一差异。[2]粒径和可压缩性会影响柱床高度的设计,因此压力-流速特性也应一并确认。[1][6]
耐清洗性与寿命
在位清洗(cleaning-in-place,CIP)使用氢氧化钠(NaOH)。据综述,琼脂糖类和聚合物类填料可用稀 NaOH(0.05~0.2 N,接触 30 分钟)进行消毒,耐碱稳定化配基可耐受 0.1~0.5 N。[1][7]重复使用次数据报道为 100~200 次以上,[1][7]但反复使用会使配基降解、流失,结合载量下降。[4][7]寿命受清洗溶液和料液的影响,因此应以实际料液进行重复循环试验来确定。[1][7]
杂质与洗脱条件
- 脱落 Protein A(leached Protein A):指配基随抗体一同被洗脱的现象,属于工艺相关杂质。[8][15]用酶联免疫吸附测定法(ELISA)测定,[8][9]并在其后的离子交换步骤中去除。[1][8]
- 洗脱 pH:洗脱需要低 pH(2.5~4),而这一条件可能引起抗体聚集。[1][10]各填料的洗脱 pH 和聚集体比例应一并考察。[1][6]
- 宿主细胞蛋白(host cell protein,HCP):相当一部分在 Protein A 步骤中被去除,但残留量因填料、料液和淋洗条件而异。[1][11]既有填料之间差异大的报道,也有差异小的报道。[3][6]
成本与法规文件
填料成本不按每升价格,而按反映结合载量、重复使用次数和年批次数的每 g 抗体成本来衡量,是合理的做法。[7][12]
确认供应商是否有法规支持文件(regulatory support file),如果正在准备美国上市申请,则确认能否取得药物主文件(Drug Master File,DMF)的引用授权书。[17][18]获批后更换填料材质属于纯化工艺变更。[14][16]ICH Q5E(2004年 Step 4,现行)认为变更前后的产品不必完全相同,但必须高度相似,并规定应以质量属性分析、杂质去除能力、工艺控制和稳定性资料证明这一点。[14][19]
